Estrone – a brief overview





What it is:


Estrone (often abbreviated as E₁) is one of the three major natural estrogens found in humans.



How it’s made:


It’s produced mainly from adrenal precursors such as dehydroepiandrosterone sulfate (DHEAS). In adults, most estrone is formed by conversion of androstenedione in peripheral tissues—especially fat tissue—and by aromatase activity in adipose and other cells.



Why it matters:


Estrone is the predominant estrogen after menopause. It’s less potent than estradiol (E₂) but still crucial for maintaining bone density, cardiovascular health, and overall hormonal balance. In conditions with high androgen levels—like polycystic ovary syndrome (PCOS)—estrone can be elevated because excess androstenedione is aromatized to estrogen.



Clinical relevance:


Knowing how estrone is produced helps clinicians assess hormone-related disorders. For example, measuring estrone levels assists in diagnosing PCOS or evaluating endocrine status after menopause, and informs decisions about hormone replacement therapy or anti-androgen treatments.

Understanding the pathway of estrone synthesis clarifies why certain conditions alter estrogen balance and guides targeted interventions for reproductive and metabolic health.
